Abstract

The global burden of malaria continues to present serious obstacles to public health, particularly in sub-Saharan Africa and parts of Asia. Traditional antimalarial drugs have been instrumental in combating malaria, but issues such as drug resistance, side effects, and suboptimal bioavailability have prompted the development of innovative drug formulations and delivery systems. Recent advancements in nanotechnology, targeted drug delivery, and controlled-release formulations have enhanced the therapeutic efficacy of antimalarial agents. Nano-carrier systems, such as liposomes, micelles, and dendrimers, offer improved drug stability, targeted action, and reduced toxicity, addressing limitations of conventional treatment regimens. Additionally, transdermal, inhalable, and medication delivery system that are implanted have been explored to improve patient adherence and optimize dosing. The integration of diagnostic tools with therapeutic delivery systems, as seen in combination therapies, allows for better management of drug-resistant malaria. The most effective method for malaria treatment is Artemisinin-based Combination Therapy (ACT) for uncomplicated cases, as it rapidly eliminates parasites and prevents resistance through dual-drug action. For severe or complicated malaria, intravenous artesunate is the gold standard, providing faster parasite clearance and significantly reducing mortality compared to older drugs like quinine. This review highlights the key advancements in the formulation and delivery systems of antimalarial drugs, focusing on the development of novel technologies and their potential to revolutionize malaria treatment, reduce transmission, and mitigate the challenges of resistance and adverse effects.
